Charles A. Gersbach's indexed work centers on programmable CRISPR-Cas9 tools for activating and silencing genes, spanning transcriptional activation, epigenetic repression, and light-inducible control of endogenous loci. His most-cited lead-author contributions include CRISPR-Cas9-based acetyltransferase epigenome editing that activates genes from promoters and enhancers (1,896 citations) and RNA-guided gene activation by Cas9-based transcription factors (1,328 citations), alongside highly specific Cas9 repressors for silencing distal regulatory elements (1,013 citations). A widely cited review of next-generation CRISPR-Cas technologies and applications (1,462 citations) frames this line of inquiry. Beyond these, his portfolio includes middle-author contributions such as ZFN, TALEN, and CRISPR/Cas-based methods for genome engineering (3,818 citations), an early comparative survey of editing platforms. Lead-author work also extends toward translation, including multiplex correction of dystrophin mutations for Duchenne muscular dystrophy and its long-term AAV-CRISPR evaluation. No software, datasets, patents, or grants are indexed beyond publications. Across these outputs, the throughline is a sustained effort to make epigenome and genome editing programmable, specific, and therapeutically viable.
