Dan R. Littman's indexed work centers on the transcriptional control of Th17 differentiation, notably identifying the orphan nuclear receptor RORγt as the driver of proinflammatory IL-17+ T helper cell programs, a finding that anchors a body of work on mucosal immunity. Related studies examine how segmented filamentous bacteria induce intestinal Th17 cells and how TGF-β-induced Foxp3 antagonizes RORγt, together mapping the balance between tolerance and inflammation at barrier surfaces. Earlier work on the fractalkine receptor CX3CR1 and on monocyte subsets helped establish how innate cells are classified and recruited. Beyond these, middle-author contributions include the identification of a major HIV-1 co-receptor and DC-SIGN as a dendritic-cell protein enhancing trans-infection of T cells, reflecting a parallel line of inquiry into host-pathogen interactions. No software, datasets, patents, or grants are indexed beyond publications. Across these works, the throughline is mechanistic immunology—linking receptor signaling, microbiota, and T-cell fate decisions—often in collaboration with researchers such as Alexander Yu Rudensky and Curtis Huttenhower.
