David E. Root's indexed work centers on functional genomics toolkits and their application to cancer biology. His senior-author contributions include optimized sgRNA design to maximize CRISPR-Cas9 activity while minimizing off-target effects (4,632 citations) and earlier rational design of highly active sgRNAs for gene inactivation, work that helped establish practical rules for loss-of-function screens. This line extends back to lentiviral RNAi libraries for arrayed, high-content screening of human and mouse genes and genome-scale loss-of-function screening, alongside a public genome-scale lentiviral ORF expression library for gain-of-function studies. Beyond these, Root contributed as a middle author to genome-scale CRISPR-Cas9 knockout screening in human cells and to systematic RNAi work showing KRAS-driven cancers require TBK1, both of which shaped how dependency screens are interpreted; collaborators on such efforts include William C. Hahn and Matthew M. Meyerson. No software, datasets, patents, or grants are indexed beyond his publications. Taken together, the arc runs from chemical-biology probe annotation and RNAi library construction toward CRISPR guide design and large-scale functional screening resources.
