David G. DeNardo's research centers on how innate and adaptive immune cells shape tumor progression and therapy response, particularly in breast and pancreatic cancer. His most-cited work includes macrophage regulation of tumor immunity and immunotherapy, a widely cited synthesis (over 2,500 citations) framing myeloid cells as therapeutic targets. Earlier studies established that leukocyte complexity predicts breast cancer survival and modulates chemotherapy response, and that CD4+ T cells promote pulmonary metastasis by enhancing protumor macrophage properties. Together these define a sustained inquiry into how myeloid–lymphoid crosstalk governs pro- versus anti-tumor immunity. In parallel, senior-author work translated these insights toward treatment: CSF1/CSF1R blockade reprogramming tumor-infiltrating macrophages to improve checkpoint immunotherapy in pancreatic models, and FAK inhibition sensitizing pancreatic cancers to checkpoint blockade. He also contributed to a framework for understanding cancer-associated fibroblasts and to a phase 1b trial of CCR2 inhibition combined with FOLFIRINOX in pancreatic cancer. No software, datasets, patents, or grants are indexed beyond these publications. The arc runs from describing immune complexity in breast cancer to therapeutically targeting myeloid populations across solid tumors.
