Caleb A. Lareau's indexed work centers on single-cell technologies for probing chromatin accessibility and mitochondrial genetics, often combining the two. His most-cited first-author studies introduce droplet-based combinatorial indexing for massive-scale single-cell chromatin accessibility and massively parallel single-cell mitochondrial DNA genotyping alongside chromatin profiling, the latter underpinning later analyses of purifying selection across human immune cells in mitochondrial DNA disorders. Earlier computational contributions include hichipper, a preprocessing pipeline for calling DNA loops from HiChIP data, and diffloop, a framework for identifying differential DNA loops. Beyond these, Lareau contributed to widely used community resources, including Signac for single-cell chromatin state analysis and the Activity-by-contact model of enhancer–promoter regulation, both middle-author roles. He also helped map the continuous regulatory landscape of human hematopoietic differentiation and, with Ansuman T. Satpathy, linked mitochondrial mutations to human lineage tracing. No software, datasets, grants, or patents beyond publications are indexed. The throughline is methodological: building tools that make single-cell chromatin and mitochondrial data interpretable, then applying them to immunology and disease.
