David Arthur Tuveson's research centers on the biology and treatment of pancreatic ductal adenocarcinoma, with a sustained focus on how oncogenic signaling and the tumor microenvironment shape disease progression. His senior-author work established that Trp53R172H and KrasG12D cooperate to drive chromosomal instability and metastatic pancreatic cancer in mice, a model that underpins much of the field's preclinical work. He later showed that Hedgehog signaling blockade improves chemotherapy delivery in pancreatic tumors, and that pancreatic cancer contains distinct populations of inflammatory fibroblasts and myofibroblasts, reframing the stroma as a heterogeneous, functionally organized compartment. Beyond these, Tuveson contributed to a cross-species single-cell analysis of pancreatic cancer revealing antigen-presenting cancer-associated fibroblasts, work that connects to David G. DeNardo's studies of stromal heterogeneity. Earlier in his career he was a co-author on the imatinib mesylate trial in advanced gastrointestinal stromal tumors, and he helped develop organoid models of human and mouse pancreatic cancer that have become widely used experimental platforms. No software, datasets, or patents appear in the indexed record; his contributions are published works spanning tumor genetics, stromal biology, and preclinical model development.
