Costas Andreas Lyssiotis studies how metabolic rewiring sustains cancer, with a particular emphasis on pancreatic tumors and their microenvironment. His most-cited lead work maps metabolic crosstalk between tumor cells and their surrounding niche, and his senior-authored review of tissue-specific and microenvironmental metabolic networks in mutant KRAS-driven tumors extends that framework across cancer types. A related strand examines how metabolism can be exploited clinically, including employing metabolic dependencies to improve pancreatic cancer diagnosis and treatment, and how macrophage-released pyrimidines blunt gemcitabine efficacy. His broader contributions include middle-author roles in foundational pancreatic cancer metabolism studies, among them work showing that oncogenic KRAS maintains tumors through anabolic glucose metabolism and that glutamine supports pancreatic cancer growth via a KRAS-regulated pathway, collaborations involving Alec C. Kimmelman and David Arthur Tuveson. Later senior-authored work identified GOT1 inhibition triggering ferroptosis and uridine-derived ribose fueling glucose-restricted pancreatic cancer. No software, datasets, patents, or grants are indexed beyond these publications, so the throughline is a sustained inquiry into how pancreatic tumors source and route nutrients.
