Koustav Pal’s indexed work centers on the computational and structural analysis of three-dimensional genome organization. Their research frequently utilizes Hi-C sequencing to explore how chromatin folds and interacts within the nucleus, particularly in the context of gene regulation. A primary focus of their lead-author contributions includes a comprehensive framework for Hi-C data analysis that spans from initial data generation to complex integration, a work that has received over 110 citations. Pal has further applied these methods to evolutionary biology, specifically identifying global chromatin conformation differences in the Drosophila dosage compensated chromosome X. This research suggests that higher-order genome architecture plays a critical role in the coordinated transcriptional up-regulation required for sex-chromosome balancing. Beyond lead-author studies, Pal has contributed to high-impact collaborative efforts, including a comparative benchmark of computational methods for Hi-C data analysis and the development of the lncRNome knowledgebase for human long noncoding RNAs. To support the technical demands of the field, they developed HiCBricks, a software tool for the efficient handling of large-scale Hi-C datasets, which facilitates the storage and retrieval of multi-resolution genomic matrices. Their more recent outputs extend into the biochemical properties of chromatin domains and the reconstruction of enhancer-target gene regulatory networks. Collectively, these contributions reflect a consistent arc of developing and applying bioinformatics tools to decode the spatial complexities of the eukaryotic genome.
